胚胎小鼠大脑半球培养神经细胞缺氧缺血后γ-氨基丁酸摄取的变化

摘要 用体外培养小鼠胚胎大脑半球神经细胞,以缺糖缺氧模拟缺血损伤,观察缺血后神经细胞GABA摄取能力的变化。结果显示,缺血3hGABA摄取明显降低,为对照组的78.8%。恢复48h后仍仅为对照组的70.8%。缺血时间越长,GABA摄取越低。说明GABA摄取能力的变化可以做为体外培养神经细胞损伤的观察指标。GABA摄取的下降可能是膜的不可逆损伤的结果。 AN Approach of evaluating the effect of anoxia-ischemia on the uptake of GABA in cerebral neuronal cells is reported.Cerebral cortices from fetal mice were utilized for preparing primary dissociated neuronal cell culture. The lack of oxygen and glucose in vitro was used to imitate ischemia in vivo. The studies showed that the GABA uptake was decreased to 78.8% and 70.8% of the control after 3 hour' ischemia and 48 hours' recovery from ischemia respectively. These data suggest that change of GABA uptake is one of the parameters of nerve cell damage. The decrease of GABA uptake may be the result of irreversible damage to cell membrane.
