摘要
1989─1992年间对杭州等地一串红(Salviasplendens)花叶病的发生及其病原进行了系统研究,从病株上分离得到的17个病毒分离物,经生物学和血清学反应测定均符合黄瓜花叶病毒(CMV)的特点。病毒分离物M-22不能经由桃蚜传播使黄苗榆等供试寄主发病。用0.5mol/L磷酸钾盐缓冲液对M-22进行提纯,提纯病毒经醋酸铀负染,测定其粒子大小为30.2nm。SDS-聚丙烯酰胺凝胶电泳测定M-22的外壳蛋白分子量约为28kD,经琼脂双扩散测定,M-22与几个已知CMV抗血清均有强阳性反应,M-22与一个番茄不孕病毒(TAV)抗血清有阳性反应,但其抗血清与供试TAV抗原无阳性反应。ELIsA反应测定结果表明,M-22与一个来自于十字花科大白菜上的CMV分离物和一个来自于前科烟草上的CMV分离物,在血清学关系上有明显差异。根据实验结果,作者认为目前在杭州等地侵染一串红的病原病毒主要是CMV,侵染一串红的CMV分离物M-22可能是一个独立的血清型.
Seventeen virus isolates were obtained from diseased plants of Salvia splendens.All the isolates were identified as cucumber mosaic virus(CMV)on the basis of biological properties,morphological and serological characteristics. One tested isolate M-22 was not transmitted by Myzus persicae in non-persistant manner. A modified schedule for purification was applied whichyielded virus 10 mg/100 g fresh tobacco leaf.One single protein species that was about 28. 0 kDwas detected by SDS polyacrylamide gel electrophoresis。When tested by gel-diffusing and ELISA,the isolate differed serologically from two CMV isolates from Brasica napus(CMV-H2)and tobac-co(CMV-8)respectively,This isolate as showed weak reaction with an antiserum of tomato as-permy virus.The isolates from S. splendens was considered as a separate serologicalgroup ofCMV.CMV was considered as the main pathogen causing mosaic disease on Salvia splendens.
