摘要
本研究用免疫印迹技术,以纯化的HSP70为抗原,检测了高温作业工人血清抗HSP70抗体的水平。实验发现;用人体来源的HSP70作为抗原的检测比动物(大鼠)来源的HSP70作为抗原的反应灵敏得多。用人体细胞制备的HSP70作为抗原时,10例高温作业工人中有2例呈阳性反应,而非高温对照10人中仅1例呈弱阳性反应,含此抗体阳性血请用动物来源的HSP70作抗原时显著减弱。用动物来源的HSP70作抗原,检测50例不同时象(以高温工人对比,白血病、心肌炎、肝癌等患者和正常对照者)的血清,除2例高温工人呈阳性反应外,余均为阴性。动物与人体来源的HSP70免疫源性差异,可能是产生不同结果的主要原因。通过本研究,我们认为用纯化的HSP70或其他HSPs作为抗原来检测人体抗HSPs抗体的方法是可行。
n this experiment,anti-HSP70 of serum in different groups
such as workers working inthe extremely hot enviroment,students , patients with leukermia ,
liver cancer and heart dis-ease were detected with Western blot using purified HSP70. The
results shows that there ex-isted a difference when using HSP70 antigen of different origins
using human HSP70 as anti-gen,2 obvious positive stainings were observed in 10 workers
working in the very hot enviro-ment and I in 10 students. Using animal-original HSP70 in 50
human sera,only 2 workersshowed as positive in workers working in the very hot enviroment.
Unfortunately,we havenot yet investigated the characters of these two individuals. Anyway, this
result offered apo.sible way for large-scale immunological investigation.
