粒细胞白血病患者环核苷酸代谢的研究 被引量:4

摘要 通过对21例粒细胞白血病患者血浆环一磷酸腺苷(cAMP)含量、腺苷酸环化酶(AC)、环腺苷酸磷酸二酯酶(PDE)比活性和环一磷酸鸟苷(cGMP)含量进行检测,并与18例非白血病患者和22例正常人血浆cAMP、cGMP含量和AC、PDE比活力进行比较,发现急性和慢性粒细胞性白血病组的cAMP水平显著低于正常对照组(P<0.01)、AC比活力也显著低于正常对照组(P<0.01)、cGMP水平显著高于正常对照组(P<0.01),而PDE比活力较正常对照组无明显差异(P>0.05)。在非白血病组,上述指标较正常对照组的差异无显著性(P>0.05)。本文结果提示粒细胞白血病患者cAMP水平降低并伴随着cGMP水平的增高,呈相反现象,而cAMP水平降低的原因系由于腺苷酸环化酶(AC)比活力下降的结果,并对AC比活力下降的机理以及造成粒细胞白血病及其前期细胞快速增殖和异常分化的病理生理机制进行了讨论。 The plasma levels of cAMP,cGMP and specific activity of adenylatecyclase (AC),cAMP-phosphodiesterase(PDE)were measured in 21 patients withacute myelogenous leukemia and chronic myelogenous leukemia. The comparisonof cAMP,AC,PDE and cGMP levels was performed on 18 nonleukemic patients and22 normal subject.We found that the plasma cAMP levels and AC specific activity weresignificantly reduced(P<0.01),cGMP levels were markedly elevated(P<0.01),whereasePDE specific activity have no marked difference from those of normal subjects(P>0.05).In nonleukemic group,the levels of cAMP,AC,PDE and cGMP had no signifi- cantly differences(P>0.05). Experimental data indicated that the reduction of plasmacAMP levels was the result of reduction of AC activity, and suggest that the reductionof plasma cAMP levels was accompanied by the eleyation of plasma cGMP levels in hu-man myelogenous leukemic patients.The mechanism of reduc tion of AC specific activityin myelogenous leukemia patients was discussed.
