花生四烯酸氧化产物对肾毒性肾炎肾脏血液动力学的影响 被引量:3

摘要 应用血栓素合成抑制剂、5-脂氧化酶抑制剂及12-脂氧化酶抑制剂对肾毒性肾炎大鼠进行预处理,测定其肾小球滤过率、肾血流量及肾小球合成的前列腺素E2、血栓素B-2、白三烯B-4及12-羟二十碳四烯酸。发现这三种抑制剂分别抑制肾小球合成血栓索B-2、白三烯B-4及12-羟二十碳四烯酸,并缓解了肾小球滤过率及肾血流量的下降程度。由此证实以上三种物质参与肾毒性肾炎中的肾脏血液动力学紊乱。 Glomerular synthesis of prostaglandin(PG) E-2, thromboxane(TX)B-2,leukotriene( LT ) B and 12-hydroxyeicosatetraenoic acid( 12-HETE )and glomerular filtration rate ( GFR ),renal blood flow (RBF)were studied in rat nephrotoxic serum nephritis( NSN ). Reduced GFR and RBF and elevated glomerular synthesis of PGE-2,TXB-2, LTB-4 and 12-HETE were found after NSN induction. The pretreatment of rats with TX synthetase inhibitor Furegrelate,50-lipoxygenase inhibitor U66858 or 12-lipoxygen-ase inhibitor Baicalein markedly ameliorated the decrements in GFR and RBF induced by anti-GBM antibody.Enhanced glomerular formation of TXB-2, LTB-4 and l2-HETE in NSN were inhibited by administration of Furegrelate, U66858 and Baicalein respectively. These data provided an evidence that TX, LT and 12-HETE mediate the perturbation of renal hemodynamics in NSN.
