苯妥英锌抗脂质过氧化及对小鼠缺血心肌的保护作用 被引量:1

摘要 本研究观察苯妥英锌(PHTZ)对异丙肾上腺素(ISO)引起小鼠急性心肌缺血的保护作用,并与苯妥英钠(PHTS)比较。结果发现,与对照组相比,缺血组血浆MDA含量升高18.2%(P<0.05);红细胞SOD活力降低27.8%(P<0.01);全血GSH-Px活力降低17.6%(P(0.01),心肌组织出现明显缺血性损伤(P<0.01)。与缺血组相比,PHTS及PHTZ组MDA含量分别降低26.3%(P<0.01)及20.2%(P<0.01);全血GSH-Px活力升高33.8%(P<0.01)及29.2%(P<0.01);PHTZ组SOD活力升高30.2%(P<0.01)。PHTS组SOD活力低于对照组(P<0.01)和PHTZ组(P<0.05),而与缺血组无差异,PHTS组GSH-Px活力高于对照组(P<0.05),PHTS组及PHTZ组心肌组织缺血损伤明显减轻(P<0.01)。结果表明PHTS与PHTZ均具有抗自由基引发的脂质过氧化反应、保护GSH-Px活力及保护缺血心肌作用,PHTZ保护SOD活力的作用可能与锌有关。 The protective effects of phenytoin zinc(PHTZ )and pbenytoin sodium(PHTS)on ischemic myocardium were investigated on experimental myocardial infarction model induced by subcutaneous injection of isoproterenol ( ISO)into mice.The results showed that(1)The MDA cont ents were increased by 18.2%(P<Q.05),SOD and GSH-Pxactivities decreased by 27.8%(P<0.01)and 17.6%(P<0. 01) respectively and severe my-ocardial ischemic lesion occurred In ischemic group,as compared with the control group;(2)MDA contents were decreased by 20.2%(P<0. 01 ) and 26.3%(P<0.01) respecti-vely,and GSH-Px activities were increased by 29.2%(P<0.01) and 33.8%(P<0.01 )respectively in PHTZ and PHTS groups, as compared with the ischemic group;(3 )Adm-inistration of PHTZ and PHTS protected myocardiam from ischemic lesion significantly;(4)SOD activities in PHTZ group were increased by 30.2%(P<0.01 ), as compared with ischemic grou.There was significant reduction of SOD in PHTS group,as com-pared with control group(P<0.01)and PHTZ group(P<0.05).The results indicated that PHTZ and PHTS possessed protective effects on blood GSH-Px activities, antilipid pero-xidation reaction and protective effects on ischemic myocardium,and that protective ef-fects of PHTZ on erythrocyte SOD activities was probably resulted from its zinc.
