大鼠免疫细胞的促肾上腺皮质激素受体的研究

摘要 大鼠免疫细胞都有高、低亲和力两类ACTH受体。脾脏细胞高亲和力促肾上腺皮质激素(ACTH)受体的解离常数(KD)为0.11nmol/L,最大结合量(Bmax)为1.2fmol/2×10 ̄6细胞,366位点/细胞,低亲和力受体KD为5.7nmol/L,Bmax为147fmol/2×10 ̄6细胞,42000位点/细胞。外周血淋巴细胞(PBL)高亲和力ACTH受体的KD值为0.15nmol/L,Bmax为1.2fmol/1×10 ̄6细胞,1580位点/细胞,低亲和力受体KD值为6.1nmol/L,Bmax为37.5fmol/1×10 ̄6细胞,32000位点/细胞。胸腺细胞、T、B淋巴细胞及腹腔巨噬细胞(Mφ)亦具有高、低亲和力不同的ACTH受体。 It was reported that rat immune cells carry both high and low affinity ACTH receptor(ACTHR)·Splenocyte, its high affinity ACTHR had 366 sites/cell,its KD=0.11nmol/L and Bmax=1.2fmol/2 × 10 ̄6 cells and low affinity ACTHR had 42000 sites/cell,its KD=5.7nmol/L and Bmax=147fmol/2×10 ̄6 cells.Peripheral blood lympho-cyte(PBL),its high affinity ACTHR had 1580 sites/cell,its KD=0.15nmol/L and Bmax=1.2fmol/1×10 ̄6 cells,and low affinity ACTHR had 32000 sites/cell, its KD=6.1 nmol/L and Bmax = 37.5fmol/1×10 ̄6 cells. Thymocyte,its high affinity ACTHR had 840 sites/cell with KD=0.15nmol/L and Bmax= 2.8fmol/2×10 ̄6 cells, and low affinity ACTHR had 12400 sites/cell with KD=4.2nmol/Land Bmax = 41.5fmol/2×10 ̄6 cells,T lymphocyte separated by nylon wool column method, its high affinity ACTHR had 66 sites/cell with KD=0.1 nmol/L and Bmax=0.22 fmol/2×10 ̄6 cells and low affinity ACTHR had 2600 sites/cell with KD = 6.1 nmol/L and Bmax = 8.8 fmol/2 × 10 ̄6 cell.B lymphocyte separated by anti-θ-serum pluS complement,its high affinity ACTHR had 1470 sites/cell with KD=0.24nmol/L and Bmax=1.7fmol/7 × 10 ̄5 cells,and low affinityACTHR had 9976 sites/cell with KD=2.4 nmol/L and Bmax = 11.6 fmol/7×10 ̄5 cells. Peritoneal adherent cells( Mφ),its high affinity ACTHR had 516 sites/csll,with KD=0.08nmol/L,and Bmax= 0.86 fmol/1×10 ̄6 cells,and low affinity ACTHR had 22000 sites/cell,with KD =10.5nmol/Land Bmax= 36.8fmol/1×10 ̄6 cells.
