山莨菪碱对内毒素血症大鼠肠系膜血管内皮细胞与白细胞粘附力的影响 被引量:15

摘要 本文采用超高速摄录相和荧光显微系统装置,检测了活体大鼠肠系膜微循环中同一微血管内的白细胞流态和血流速度的变化,观察了山莨菪碱对内毒素作用下的微静脉内皮细胞与白细胞粘附反应增强的影响。结果显示:内毒素血症大鼠的微动、静脉口径、微动脉血流速度不变,微静脉血流速度后期明显降低,30min下降16.67%(P<0.05);微静脉内附壁滚动和粘附的白细胞数量迅速增多,粘附密度指数上升;白细胞与微静脉内皮细胞间的粘着系数(adhesioncoefficiency,AC)升高16.34~46.82%左右(P<0.01)。山莨菪碱表面滴注能拮抗内毒素对白细胞流态和性质的干扰,明显减少白细胞附壁滚动和粘附数量,降低白细胞与内皮细胞间的粘着系数,加速微循环流动;但对正常大鼠微静脉内白细胞的流态和血流速度无明显作用。由此提示,山莨菪碱主要是通过降低被内毒素激活的白细胞与内皮细胞间的粘附力、减少白细胞释放生物活性物质,来保护血管内皮细胞和改善休克时的微循环血流障碍。 The effect of anisodamine on neutrophil-mediated reactivity of mesentericvenular endothelium was studied in experiment endotoxemia by using fluoresent micros-cope equipped with a SIT image tube camera and high speed recording TV system.Theleukocytes on venule were visualized by injection of acridine orange. Endotoxin increasethe number of rolling and adherent leukocyte on venular endothelium rapidly,the den-sity index of adherent leukocyte was also elevated significantly. The adhesion coefficiencybetween leukocyte and venular endothelium was increased 27.15%and 46.82%at 15'and 30 'of endotoxin infusion respectively.Anisodamine topical application can antag-onize the damage effect of endotoxin,lessen rolling and adherent leukccytes number ,dropadhesion coefficiency between leukocyte and endothelium in venules,and improve micro-circulatory blood flow. But it dosen't affect normal animal. These suggest that endo-toxin can change the behavior of leukocyte, while the anti-shcck effect of anisodamineis mediated by its ability to reduce adhesion between leukocyte and venular endotheliumin microcirculation.The improvement of blood flow velocity may be the result of reliefof microcirculatory disorde
