高温、CO及其联合作用对大鼠HSP70mRNA,HSP70合成影响研究 被引量:2

摘要 当组织细胞对热或其它刺激反应时,均被诱导增加HSPs合成。本研究将8只雄性大鼠随机分为四组,分别为对照组、高温组、CO组、高温+CO组。在送氧式高温与毒物联合作用装置中染毒1h,在室温下恢复1h后,测定其肛温、HbCO。断头处死,取其肝脏分析HSP70mRNA和HSP70。结果如下:(1)与对照组相比,高温组、高温+CO组大鼠肛温均升高3.0℃以上,CO组及高温+CO组大鼠HbCO含量均达30%以上,这说明受热及染毒后动物的体温及HbCO含量均符合本实验要求。(2)结果显示,对照组仅有少量HSP70mRNA,与之相比,CO组有所增加,高温组增加明显,而高温+CO组增加更为明显,表现为协同作用。HSP70合成也与HSP70mRNA结果一致。 he HSPs(heat stress proteins)could beinduced by heat or other stimuli,the normalproteins was inhibited. Present study aimedat the effects of high temperature,carbonmonoxide and their combined effects on the major heat stress protein-HSP70 and its mRN-A.8 male rats were randomly divided intofour groups: control group(25.0+1.0℃, hightemperature group(40.0 + 2.0℃),carbon mon-oxide group(600+50ppm, 25.0+1.0℃)and thecombine effects group(600+50ppm, 40+2.0℃).After experiment, the rats rceovered in roomtemperature for 60 min. Then determined therectal temperature and HbCO in the blood,the liver were taken for HSP70 and its mR-NA for analysis. The results were as follws:(1)the rectal temperature in high tempera-ture group and combined effect group, show-ed a 3. 0℃;HbCO level in CO group andcombined effect group increased above 30%,respectively compared with control group.(2)only little HSP70 mRNA was transcripted in control group, showed little increase HSP70mRNA was obviously increased in high temp-erature group, and the increase of HSP70mRNA in combined effect group was most obvious, suggesting a synergistic action betw-een high temperature and CO. The synthesisof HSP70 showed a consistent changes withthat of its mRNA in all groups.
