刚地弓形虫细胞培养的研究Ⅳ.临床标本培养检测技术的改进

摘要 本文以血清抗弓形虫抗体阳性的孕妇羊水进行细胞培养检测,实验组以THP一1细胞系培养并加入经离心过滤的RH株速殖子细胞培养上清(SRC)作为培养佐剂,在第4~6d,第7~10d,第11~14d进行3次检验;对照组以MRC5细胞系培养,在第4d、第8d进行2次常规检测。结果26例羊水在MRC5组全部阴性,而THP一1组分离出3株野生株,并在其后经同时接种小鼠的血清特异性IgM抗体和脑包囊检测或脐血特异性IgM检测所证实。提示加用SRC并延长培养检测时间对提高弓形虫病以细胞培养作病原诊断的敏感性有显著意义。 Twenty一six specimens of amniotic fluid from pregnant women were cultured withTHP一1or MRC5 cell line,of which the former as experimental group cultured together with super-natant of medium of RH strain culture(SRC)and detected on day 4一6,day 7一10, day11一 14,re-spectively,the latter cultured without SRC and detected two times on day 4,day 8 respectively as theroutine method. The results showed that in THP一1group three positive cases were found, the posi-tive rate was 11.5%,while in MRC5 group all negative,The results were verified by cyst detectionin mice inoculated with 2 positive specimens and by specific IgM and IgA detection of umbilical and fe-tal sera from another case. The results indicated that the technical improvement in adding SRC andprolonging the culture/detection time could remarkably increase the sensitivity of pathogenic diagnosisby means of cell culture.
