采用混菌生检方法作妥布霉素产生菌诱变预筛选

摘要 以对妥布霉素和卡那霉素高度耐药而对阿泊拉霉素敏感的金葡球菌11-R7和对3种抗生素都敏感的枯草杆菌63501为检定菌,建立了能同时检测尼拉霉素(nebramycin)复合物总效价和组分相对含量的预筛选方法。并据此比较了不同处理条件对黑暗链霉素菌的诱变效果,亚硝基胍pH8处理优于亚硝基胍pH6处理和紫外线照射。孢子悬液应及时用于筛选,以免放置过程中性状的不利变化。 Two different test organisms,Staphylococcus attretts 11-R7 which washighly resistant to tobramycin and kanamycin but remained sensitive to apramycin andBacillus sutilis 63501 which was sensitive to all of the above-mentioned antibiotics were usedin one plate to assay simultaneously the total titres as well as the relative contents ofnebramycin complex for pre-selection and mutation studies. Mutation effects of differenttreatments on Streptomyces tenebrarius were compared on the basis of the pre-selectionresults. Treatment with NTG at pH8 was better than those of treatment with NTG at pH6and with ultraviolet light. Spore soltition should be used immediately after mutationtreatment to avoid adverse variations of strain characters。
