摘要
胰酶水解绵羊红细胞(SRBC)膜得到粗提物,经离子交换层析部分纯化后,获得组分TRF-Ⅲ(Trypsin-releasedfraetionⅢ)具有明显抑制SRBC与猪外周血淋巴细胞形成E玫瑰花的活佐,花环形成率与TRF-Ⅲ含量有明显的函数关系。电泳测定其分子量为66kD。TRF-Ⅲ单独作用尽管对PBMNC的增殖无作用,但可增强PHA刺激PBMNC的增殖作用,TRF-Ⅲ单独作用于PBMNC可显著增强后者的促凝血活性,与正常相比,促凝率约增加30%,与PHA促凝率(33%)相似。结果提示,TRF-Ⅲ具有E受体配体促淋巴细胞激活作用。
he protein released from intact sheep erythrocytes
by incubation with trypsin was purifiedwith DEAE-cellulosei
on-exchange chromatography. The trypsin-released fraction(TRF-
Ⅲ)containd activity of bingding to T lymphocytes and produced
inhibition of E rosette formation. SDS-PAGE and nondena ture PAGE
analyses showed a single band with MW of about 18 000and 66 000
respectively in the final purified TRF-Ⅲ.In proliferation tests,the
results showedthat the combinations of TRF-Ⅲ and submitogenic
concentration of PHA significantly stimtilat-ed proliferation of
PBMNC while TRF-Ⅲ alone was nonmitogenic. Contrast with
requirementsin PBMNC proliferation experiment,TRF-Ⅲ alone in the
procoagluation experiment similar tosheep erythrocytes increased the
procoaglutation time by about 30%.These results suggest thatTRF-Ⅲ
contains the activity of E receptor liatural ligand.
