摘要
从5株抗HLA-A2单抗杂交瘤细胞株中提取mRNA,以特异性引物采用PCR技术,扩增和克隆出单抗可变区重链基因,并采用双脱氧链式终止法及计算机基因文库测定和分析了其全部的氨基酸序列,对抗体的生物学特性的研究具有重要意义,为构建基因工程抗体奠定了基础。

characterized and determined the V region gene sequences of a series
of mousemonoclonal anti-HLA-A2. Total cellulsr RNA was isolated from
hybridoma cell lines producingthe five monoclonal anti-HLA-A2.
Cloning and sequencing of cDNA from these antibodies. Wehave aligned
them to evaiuate V region of structures homology.HO-1, HO-3, HO-4 and
HO-5express V_H regions which are highly homologous to the Vn J558
gene family and use members ofthe Dsp2. 2 D region gene and utilize
the Jx3. Alternatively,HO-2 is related to the Vn Q52 genefamily and
expresses of the DFL16. 2 and D gene regions in conjunction with the
Jx4 gene seg-ment. These date suggest that the antigen binding
specificity of an immunoglobulin molecule isdetermined by the amino
acid sequence of the variable region of heavy and light chaiii. The
heavvchains play a major role in determining the specificities of the
antibody combining sites epitoperecognition among the monoclonal
antibodies may reflect sequence variability primarily withinthe CDR3
region of Vu.
