摘要
研究了以产生抗人小细胞肺癌相关抗原和CD3双特异性抗体(Bispecificantibody)的杂交杂交瘤(Hybrid-hybridoma)的方法及双特异性抗体的性质。两个具有不同表型(HAT敏感/Neo拮抗和HAT拮抗/Neo敏感)杂交瘤细胞系,经常规方法进行融合。最后选出7个分泌双特异性抗体的杂交。杂交瘤细胞系并对其中一个克隆系细胞进行了测示研究。结果表明,该克隆系细胞培养上清能有效增强LAK细胞对靶瘤细胞的杀伤活性,表明其具有临床应用前景。

forthe preparation hybrid hybridoma which produce bis-pecific
anti-CD3 X anti-SCLC antibody.This is achieved by fusing two
hybridoma cell lines thatare phenotypically distinct(HAT ̄s/Neo ̄r
and HAT ̄r/Neo ̄s)and there by allow for the selection ofthe
appropriate cells,We have isolated several stable hybrid hybridomas
in which secreted bispe-cific antibodies that one specificity is
against CD3 and one against small cell lung carcinoma-asso-ciated
antigen. One of them,coded as WST-H7,showjng a qaudroma was done for
the prelimi-iiary characterizations。We show that cuIture supernatant
derived from WST-H7 can efficientlyinduce tumor-cells lysis by LAK
cells. The results suggest that WST-H7 has a potential use
astherapeutic agents for the treatment of cancer.
