亲和层析法纯化肾综合征出血热病毒大鼠单克隆抗体

PUKIFICATION OF RAT MONOCLONAL ANTIBODIES AGAINSTHEMORRHAGIC FEVER WITH RENAL SYNDROME VIRUS BYIMMUNOAFFINITY CHROMATOGRAPHY

应用小鼠抗大鼠Ｋａｐｐａ轻链单克隆抗体（ＭｃＡｂ）亲和层析系统，对抗肾综合征出血热病毒（ＨＦＲＳＶ）大鼠ＭｃＡｂ进行了纯化研究。使用该系统具有ＭｃＡｂ回收率好，纯度高，能较好地保持ＭｃＡｂ免疫学活性，操作简便、快速，可一步除去腹水中宿主杂蛋白，并能适应不同类别ＭｃＡｂ纯化的特点。此外还对自制盐析纯大鼠Ｋａｐｐａ轻链ＭｃＡｂ柱与进口亲和纯大鼠Ｋａｐｐａ轻链ＭｃＡｂ往进行了比较和探讨。

An affinity chromatography column was prepared by binding mouse monoclonal antibody(McAb)Mark-3 against Lou/c rat IgK-1a kappa light chain to activated sepharose 4B and theserat McAbs against HFRS virus from ascitic fluid of Lou/c IgK-1b rat were purified by this col-umn, The results showed that the recovery rates of protein of the prepared column was 14%～30%for rat McAbs ascites, Immune activity of McAbs after the purification was slightly lowerthan before. The findings demonstrated that the affinity chromatography technique of Mark-3McAb is specific,efficient,rapid and simple. It has universa applicability for different McAb sub-claes, These rat McAbs against HFRS virus purified will contribute to studies for diagnosis andimmuno-therapy of HFRS.
