摘要
用亲和层析术从孕妇尿中提取尿调素,发现:(1)尿调素浓度>100mg/L时能促进中性粒细胞对白色念珠菌的吞噬作用;(2)抑制分离的T淋巴细胞的转化,但对未分离的T淋巴细胞的转化及分泌IL-2、IFN-γ的功能无影响;(3)对SRBC免疫的小鼠脾细胞和分泌CMV单抗的杂交瘤细胞分泌抗体的功能无影响;(4)对补体经典途径的激活无影响。
romodulin(UrM)is an 85kD acidic
glycoprotein purified from the urine of normal preg-nant women,its
genes localizes to 16P13.11. Recent studies demostrated that it can
bind withcytokines and inhibit an antigen-specific human T-cell
proliferation. So it is considered as a pos-sible immunosuppressant.
The studies were aimed to probe into effect of UrM on
polymorphonu-clear leukocytes(PMNL)、 lymphocyte and complement in
vitro. Found:(1)UrM promoted thephagocytotic ability of PMNL when the
concentration was more than 100mg/L;(2)UrM inhib-ited
antigen-specific T-cell proliferation when the T-cell was isolated
from the whole blood,oth-erwise it couldn’t affect the function of
T-cell secreting IL-2、IFN-γ.(3)UrM had no effect onactivation of
classic complement pathway and on antibody-secreting cells function
includingspleen cells of mouse immunized with SRBC and hybridoma
cells secreting anti-CMV monoclonalantibody. These results suggested
UrM has no up and down regulation in vitro.
