多聚酶链反应和病毒分离检测巨细胞病毒的比较研究

Comparison of Polymerase Chain Reaction and Virus Isolation for Detection of Human Cytomegalovirus

92名住院患儿病毒分离、常规PCR、热启动PCR、碘化钠抽提结合热启动PCR的CMV检出率分别为61％、28％、39％和48％。与病毒分离止较,常规PCR、热启动PCR、碘化钠抽提结合热启动PCR检测极低拷贝数CMVDNA的敏感性分别为8％、36％和64％。从而显示碘化钠抽提结合热启动PCR,敏感性高,简便快速,4h内可判定结果,颇具临床实用价值。

Human cytomegalovirus positive rates in 92 hospitalized pediatric patients by virus isolation, conventional PCR.Hot Start PCR. Hot Start PCR with sodium iodine extraction were 61% , 28% ,39%,and 48% respectively. Compared with virus isolation .the sensitivities of conventional PCR.Hot Start PCR,Hot Start with sodium iodine extraction for extremely low-copy-number specimens were 8% ,36% and 64% repectively. The results showed that the sodium iodine extraction with Hot Start PCR was sensitive, rapid, and simple, requiring only 4 hours, therefore,it should be of high practical value in clinical application.
