Interference of SDS to Limulus Test

人基因工程IL－2在分离提取过程中需加一定量的十二烷基硫酸钠（SDS）。我们在应用鲎试验检测制剂中的热原时发现，当样本中SDS浓度＞0．025mg／ml时，可抑制自试剂的酶反应，使阳性结果转变为阴性。据此，我们建议，凡生物制剂中含有SDS或其它酶抑制剂的产品，不宜采用鲎试验检查内毒素含量。

Addition of certain quantity of SDS was necessary during the separation of recombinant IL-2. During examination of pyrogen in rIL-2 by limulus test, authors found that when the SDS concentration was ＞0．025ng/ml could inhibite the enzymatic reaction of limulus reagent resulting in comersion of positive result to negative. It suggested that limulus test should not be used for examinition of endotoxin content in the biological products containing SDS or other enzyme inhibitors.
