摘要
应用免疫荧光抗体技术检测了整合有猪瘟病毒反义基因的PK-15细胞克隆对猪瘟病毒的抑制效应。结果表明,5个不同的反义基因片段对猪瘟病毒的抑制效率存在很大的差异,其中A片段的抑制效率最高(94%~98%),B片段次之(58%~76%),C片段再次(64%以下),D片段和E片段未见明显的抑制效应。抑制效率的差异可能与反义基因片段的位置、长短以及反义RNA表达质粒载体有关。
The specific inhibition of hog chloera virus(HCV)replication by anti-sense RNA expressed in cells transfected with HCV antisense gene constructs wasdemonstrated by immunofluorescent assay(IFA)at the presence of Zn2+(50 or 65μmol/L)as inducer of metallothionein-I promoter.The inhibition rate in mMT/HCA,mMT/HCB and mMT/HCC(using mouse MT-I)transfected cell clones were 94%- 98%,58%-76%and-64%respectively.No apparent inhibition was observed in pMT/HCD and pMT/HCE (using ovine MT-I)transfected clones.The reasons for the differ-ences in inhibition effects may be resulted from the sizes and positions of the HCV anti-sense gene fragments or the as RNA expressing vectors.
