非胰岛素依赖型糖尿病患者红细胞中游离嘌呤核苷酸含量的研究

摘要 人红细胞中游离嘌呤核苷酸水平是估价非胰岛素依赖型糖尿病(NIDDM)患者红细胞能量代谢过程的重要参数。本文采用反相高效液相色谱法(reverse-phasehighperformanceliquidchromatography,rHPLC)测定NIDDM患者22例和健康人13例红细胞液中ATP、ADP、AMP和GTP的含量。红细胞经10%HClO4处理,离心后收集上清液进样。本法灵敏度可靠、特异性高。研究发现:NIDDM病人的ATP/AMP比正常人低70%,而HbA1c。为正常人的两倍,ATP/AMP的比值与HbA1c呈正相关(r=0.546)。提示测定红细胞的腺苷酸和HbA1c含量是研究糖尿病病人糖代谢和能量代谢紊乱的有效方法。 Taken the levels of free purine nucleotides in human erythrocyte as an important parameter for assessing the erythrocyte energy metabolism in patients with non-insulin-dependent-diabetes-mellitus (NIDDM) ,a high-performance liquid chromatography method was adopted for the determination of the contents of ATP,ADP, AMP and GTP in erythrocyte cytoplasma from NIDDM and normal subjects.The mean value of ATP/AMP for 13 normal subjects was about 70% higher than that in 22 NIDDM patients. The mean value of HbA1c for 22 NIDDM patients was twice as much as that for 13 normal persons.There was a positive correlation between the ATP decrease and the HbA1c increase(r=0.546).Thus analysis of nucleotides and HbA1c in human erythrocytes is a useful way for studying carbohydrate and energy metabolic disturbances in diabetes.
