鲩鱼胆汁中毒致多器官衰竭及其与氧自由基关系的探讨 被引量:7

Multiple organ failure induced by grass carp bile andthe mechanism of oxygen free radicals:experimentalstudy

摘要 用新鲜鲩鱼胆汁灌胃诱发大鼠及小鼠的多器官衰竭(MOF),探讨其与氧自由基的关系。实验结果:新鲜鲩鱼胆汁按比重法测其重量,小鼠LD_(50)为4320mg/kg体重;大鼠按3500~4000mg/kg体重灌胃,115只大鼠染毒后24~120h存活的58只中出现MOF率为77.59%;小鼠染毒后24~48h,心、肝、肾、肺、脑及血浆内丙二醛(MDA)含量明显高于对照组;染毒后24h血液内谷胱甘肽-过氧化物酶(GSH-Px)活性明显高于对照组,48h降至正常;红细胞超氧化物歧化酶(SOD)活性在48h明显低于对照组;血液内还原型谷胱甘肽(GSH)在染毒后1~6天呈3次抛物线变化趋势,染毒后1天升高,5天达最低值,6天开始回升,并逐渐恢复至染毒前水平。提示:用新鲜鲩鱼胆汁可制造MOF模型,MOF发生机制可能与氧自由基增多及抗氧化物质减少有关。 The fresh bile of grass carps was perfused into thestomaches of the rats and mice to induce multple organfailure (MOF), and the mechanism of oxygen free radi-cals in MOF was studied.The results showed that the LD_(50) in mice was4320 mg/kg.b.w. of fresh bile. Of the 115 rats poisonedby fresh bile in the dose of 3500~4000 mg/kg.b. w.only 58 remained alive for 24~120 hrs.The incidence ofMOF was 77.6g%(45/58)in the living animals.Themalonic dialdehyde (MDA ) contents in the tissues of thelivers, hearts, lungs, kidneys, brains and plasma of thepoisoned mice were significantly higher at 24~48 hrs af-ter intoxication than those of the controls. The glu-tathione peroxidase acthity was increased at 24 hrs afterbeing poisoned and returned to mormal at 48 hrs. Theerythrocyte superoxide dismutase(SOD)activity wassignificantly lower at 48 hrs after grass carp bile poison-ing than the controls. The reducing glutathione level inthe whole blood of the poisoned rats showed the tenden-cy of cubic change (y=34.15+ 6.046719x-3.635025x ̄2+0.410836lx ̄3)1-6 days after being poisoned and be-gan to increase on the first day of poisoning. The lowestlevel was seen on the 5th day and a tendency of linearascent(y= 23.34+0.82x) appeared on the 6th day afterpoisoning, thereafter the glutathione level gradually re-turned to normal. The results indicated that a new modelof MOF induced by fresh bile of grass carps was success-fully established. The multiple organ failure appeared tobe correlated with the increase of oxygen free radicalsand decrease of antioxidants in the blood and tissues.
