摘要
原代培养的人胎肝细胞中很容易混杂成纤维细胞,甚至取而代之,故而在人胎肝细胞培养中选择性地抑制、清除成纤维细胞是培养成功与否的关键。我们使用组织培养常用药品,根据细胞生长的不同状况,采取自然沉降或低速离心、部分消化与完全消化等方法,在消除成纤维细胞、纯化胎肝细胞中取得了较为满意的效果;且操作方便,易于推广应用。
Human fetal hepatocytes in primary culture are easily mixed with the fibroblasts which replace even the hepatocytes. Thus, selectively inhibiting and eliminating fibroblasts is the key to primary culture of human fetal hepatocytes. With the reagent most in use in thd tissue culture,the methods of natural precipitation, low speed centrifugation, partial dispersion and complete dispersion were used according to the different conditions of growth of the cells. More satisfactory results were achieved in our experiment of the elimination of the fibroblasts and the purification of the hepatocytes.
