猪心肌缺血再灌注时冠状静脉窦血中内皮素的变化和大剂量维生素C对其影响 被引量:3

EFFECTS OF VITAMIN C ON ENDOTHELIN CONTENTS IN PLASMA AND MYOCARDDIIUM IN EXPERIMENTAL MYOCARDIAL ISCHEMIA-REPERFUSION IN PIGS

摘要 开胸结扎猪的冠状动脉前降支60分钟,再灌注120分钟。分别在结扎前、缺血60分钟和再灌注120分钟时,从冠状静脉窦取血测定血浆内皮素(ET)的浓度,并于再灌注120分钟时取梗死心肌组织测定ET含量。结果显示,缺血60分钟时ET的血浆浓度明显升高,再灌注期进一步升高。如于缺血期推入大剂量维生素C,可显著降低再灌注期冠状静脉窦中血浆ET的升高幅度。 Left anterior descending branches of the coronary arteries in 14 pigs were ligated for 60 min followed by reperfusion for 120 min.7 pigs were given Vit C(200 mg/kg)intravenously at ischemia 10 min(Vit C group )and the other 7 pigs(control group)were given saline at the same time.Blood samples were withdrawn from the coronary sinus before ischemia,ischemia at 60 min and reperfusion at 120 min for the measurement of plasma endothelin(ET).Myocardial tissues were taken from the necrotic and normal zones of the left ventricle at 120 min of reperfusion for the measurements of ET and protein. The results showed that there was increase in plasma ET levels at ischemia 60 min(p<0. 001 vs.pre-ischemia levels) and further increase at reperfusion 120 min(p<0. 01vs. ischemia 60 min levels),in the control group,while similar trends were found in the Vit C group, but with p<0.01 and p<0.05,respectively.Vitamin C could suppress significantly the increase in plamsa ET levels at 120 min of reperfusion(p<0. 02 vs.control).No significant differences were found in the myocardial ET contents between normal zones of the two groups as well as between the necrotic zones, however,myocardial ET contents of the necrotic zones were significantly higher than those of the corresponding normal zones.The results suggest that(1)the increase of plasma ET levels might closely related to the pathophysiology of ischemia/reperfusion;(2)Vit C might play a role in the prevention of the myocardium from being damaged by the free radicals during reperfusion.
