摘要
用胞嘧啶单核苷酸酶(CMP酶)作为溶酶体标志物和视频图像数字分析仪观察大鼠腹主动脉结扎后4周、10周的肥大心肌细胞的自体吞噬泡变化。实验发现:①自体吞噬泡内有CMP酶反应产物出现;②在10周组,单位面积(24.77μm ̄2)胞质内CMP酶反应产物面积和T管面积明显大于对照组及4周组;③在10周组最明显特征是细胞降解产物排到扩张T管。这些结果说明,细胞成份的自体吞噬降解在心肌肥大过程中起着重要调节作用,扩张的T管也反映了心肌细胞外吐加快的适应性改变。
Cytidine Monophosphatase (CMPase)as a marker of lysosome and video image digital analysis system (VIDAS)were used to study changes of autophagic vacuole in the rat hypertrophied myocardium at 4 and 10 weeks after ligation of the abdominal aorta. It was found that (1)autophagic vacuole contained CMP(ase) reactive product,(2)area of CMP(ase) reactive product and area of Ttubule in unit cytoplasmic area (24.77 μm2)were significantly greater in the 10 weeks (n=6) than in the 4 weeks (n=4) and control (n=5) groups, (3) the most striking feature was expelling of degraded cellular elements into the dilated T-tubules at 10 weeks after ligation.These results suggest that autophagy of cellular elements seems to play an important regulative role in hypertrophied process and dilated T-tubules actually reflect myocardial adaptive changes.
