摘要
采用钙荧光探针Fura-2定量分析的方法研究了大黄有效成分番泻甙和大黄多糖对血小板细胞内游离钙浓度的影响。结果表明,血小板细胞在静息状态下胞浆内[Ca2+]i为142.09±17.69nmol.L-1,当依次加入CaCl2和凝血酶后,血小板细胞内游离钙浓度[Ca2+]i均明显高于静息时的水平(P<0.01)。预先向血小板悬液中加入番泻甙8×10-5~4×10-1g或大黄多糖1×10-1g后,再依次加入CaCl2和凝血酶,此时血小板细胞内游离钙浓度较对照组明显降低(P<0.01),其抑制效应与剂量相关。提示大黄及大黄制剂抑制血小板细胞聚集与番泻甙和大黄多糖抑制钙内流有关。
The effects of sennoside and RPP (Polysaccharide from rheum palmatum) on the cytoplasmic free calcium in the platelets of rats were studied by means of fluorescent probe (Fura-2) quantitative The project supported by National Natureffeence Foundation of Chiinaanalysis. The resting free Ca2+ level was 142.9±17.69 nmol·L-1 in the platelets,After adminstration of CaCl2 (2 mmol·L-1)and thrombin (500 U·L-1) sequentially.the free Ca2+ levels obviously elevated (P<0. 01),compared with the resting level ,After administration of sennoside and RPP previously. the free Ca2+ levels significantlydreased (P<0.01).conpared with thatof the control grop.Furthermore. the effects of drugs were dose-dependent.Therefore. it suggeststhat the effect of rhubarb and rhubarb preparation on inhibiting aggregation of platelets is related to sennoside and RPP.which can inhibit influx of Ca2+.
