摘要
用Percoll密度梯度离心法分离大鼠肾皮质远曲小管细胞(DTC)和近曲小管细胞(PTC).应用放射性同位素 ̄(45)Ca示踪技术观察川芎嗪对Ca ̄(2+)跨膜转运的影响,并且采用蛋白质竞争结合法检测了胞膜腺苷酸环化酶活性及细胞内环磷酸腺苷(cAMP)含量。结果表明川芎嗪明显抑制PTC和DTC细胞 ̄(45)Ca的摄取,但对 ̄(45)Ca的外排无明显影响。川芎嗪对PTC和OTC胞膜腺苷酸环化酶活性无影响,但使细胞内cAMP含量显著降低。结果提示川芎嗪影响肾小管细胞钙的摄取,其机理可能与其诱导其它物质使细胞内CAMP水平降低有关。
In order to know howtetramethylpyrazine(TMP)acts on renal calcium trans-port,proximal tubular cells(PTC) and distal tubularcells(DTC)were separated by using Percoll's gradientmethod. 45Ca was used to study the influence of TMPon calcium transmembrane. It was found that TMPsignificantly reduced the 45Ca uptake of PTC and DTC,but did not influence the 45Ca effiux. TMP also reducedthe cAMP content in PTC and DTC.but did not influ-ence the activity of adenylcyclase.The results suggestthat TMP takes part in regulating the Ca2+ transportof PTC and DTC and its effect may be related to the re-duction of cAMP content in the cells.
