摘要
建立了人血清中头孢他啶的高效液相色诸测定方法。采用C_(18)柱并用离子对试剂对其进行预处理,使头孢他啶在柱上有合适的保留时间。取50μl血清用乙腈沉淀蛋白后直接进样。线性范围0.5~400μg·ml ̄(-1)(n=7,r=0.9999,P<0.01),日间和日内误差分别低于5%和2%。具有灵敏,准确,快速,取样量和进样量小等优点,可以满足临床药代动力学和药效学研究的需要。
Reverse-phase highperformance liquid chromatographic method with C_(18)column hasbeen developed for the determination of ceftazidime in human serum. The rapid method involvedprecipitation of protein from 50 μl serum by means of acetonitrile followed by injection of super-natant directly. The linearity of ceftazidime was from 0. 5 to 400μg·ml ̄(-1)(n=7,r=0. 9999,P<0.01),and the validation of inter-day and intra-day was less than 2%and 5%,respectively. Thedetection limit was 0.5μg·ml ̄(-1).The method is sensitive and accurate;no time-consuming sam-ple preparation is required. It was special for the studies of pharmacokinetics and pharmacodyna-mics.
