摘要
本实验以醋醚铵-甲醇为流动相,硝苯地平为内标,在Hitachi3053色谱柱上对丙吡胺建立一适合临床血药浓度监测的高效液相色谱测定法。由于应用了UFC_3LGC限外滤过,使本方法更简便、快速。结果证明,该法灵敏、重现性好(r=0.9994)。考虑α_1-酸性糖蛋白(AAG)病态时浓度的变动及对药物结合的影响,对其与药物游离浓度的关系进行了分析。
We used 0.2 mol/L ammonium acetate-methanol(30:70)as mobile phase andnifidipine as internal standard to develop a HPLC method for the determination of free disopyra-mide concentration in plasma with Hitachi 3053 column. The use of UFC_3LGC made this methodmore simple and quick.The method was demonstrated to have high sensitivity and accuracy(r=0. 9994).To consider the binding of α_1-acid glycoprotein(AAG)to the drugs,we analysed for therelationship between AAG and unbond plasma disopyramide concentration.
