摘要
利用逆转录病毒介导的基因转移技术,将重组TNF-α基因导入小鼠平滑肌肉瘤细胞系S180中,对转导后肿瘤细胞的增殖、致瘤效应等进行体外和体内的观察。结果表明;逆转录病毒中介的重组TNF-α基因导入,在小鼠体外和体内都有显著的抗肿瘤效应,这为TNF-α的肿瘤基因治疗的临床应用奠定了基础。
NF-αgene was expressed in murine S180
cells(BALB/c ) by retroviral gene transfer.The trans-ferred S180
cells were inoculated into BALB/c mice s.c. to observe the
tumorigenicity in vivo and in vit-ro. In 50 days, all the five mice
inoculated with S180 cells developed tumors , while only one out of
fivemice inoculated with TNF-αgene transferred S180 cells developed
tumor and in it tumor manifestationwas significantly delayed. The
results showed that retrovirus-mediated TNF-αgene transfer
suppressedtumorigenicity of S180 cells in vivo significantly,which
were the basis for its clinical application of genetherapy for
cancer.
