摘要 应用改进的FISH(FluorescenceInSituHybridization,FISH)前Q显带方法基因定位,获得良好的Q显带核型和FISH信号,结合共焦激光扫描显微镜MRC-600系统记录杂交信号和染色体图像,经Comos系统软件处理,提高了信号的分辨率和敏感性,首次将人类内α胰蛋白酶抑制因子2.5kb小片段cDNA定位于3P21。 luorescence in situ hybridlzation(FISH ) was carried out on metaphase and pre-metaphase chro-mosomes treated by a modified Q-banding, The hybridized signal and chromosomal image were recordedby confocal laser scanning microscope(MRC- 600 ) and processed with COMOS software. The resolu-tion and sensitivity of signal were improved.We had successfully applied the modified procedure to lo-calize the inter α trypsin inhibitor gene on chromasome 3p21 using a 2. 5kb cDNA fragment of the gene.
