摘要
应用SV-40导入法建立了人骨髓基质细胞株SC5-7,然后收集其培养上清,测定其中三种造血因子含量,并将这种培养上清添加至造血干细胞的液体培养基内,每周一次取出培养的细胞,进行计数。同时移至甲基纤维素培养基内继续培养,14日后,在倒置显微镜下观察并计数各种造血干细胞集落形成的数量。结果:1.人骨髓基质细胞株SC5-7的培养上清中含有较高浓度的G-CSF、GM-CSF和M-CSF。2.添加SC5-7培养上清的培养系,比没有添加SC5-7培养上清的培养系造血干细胞体外增殖时间明显延长。结果表明:人骨髓基质细胞株SC5-7具有分泌造血因子的作用,而且在体外可以促进造血干细胞的增殖。
arrow stromal cell and its secretion
of hematopoietic factors play an important role in the prolifer-ation
and differentiation of marrow hematopoietic stem cell.To survey this
function,we build marrowstromal cell lines SC5-7 by inputting SV- 40,
then collect its culture supernate,measure the quantity ofthree
hematopoietic factors in it,and put this culture supernate into the
liquid culture medium ofhematopoietic stem cell,then take the culture
cell to count it once a week,meanwhile remove it to
themethylcellulose culture medium to continue to culture. After 14
days,observe and count the colonynumbers of all kinds of
hematopoietic stem cell.Results:1.There are higher consistency of
G-CSF,GM-CSF and M-CSF in the culture supernate of human marrow
stromal cell lines SC5-7.2.The proliferationtime of the hematopoietic
stem cell in vitro of the culture medium with SC5-7 culture supernate
is evi-dently longer than that of the culture medium without
SC5-7.This result suggests that human marrowstromal cell lines SC5-7
has the fonction of secreting hematopoietic factors,and promoting the
prolifera-tion of the hematopoietic stem cell in vitro.
