罗格列酮和白细胞介素-6对3T3-L1脂肪细胞chemerin mRNA表达的影响 被引量:1

摘要 目的 观察罗格列酮和白细胞介素-6(IL-6)对3T3-L1脂肪细胞chemerin mRNA表达的影响.方法 体外培养3T3-L1前体脂肪细胞并诱导其分化成熟,分别以不同浓度的罗格列酮和(或)IL-6干预,在不同的时间段提取细胞总RNA,采用RT-PCR技术检测细胞中chemerin mRNA的表达水平.结果 IL-6作用于分化成熟的3T3-L1脂肪细胞能下调chemerin mRNA的表达,且用罗格列酮预处理或与IL-6共同作能够改善IL-6对chemerin RNA的抑制作用.结论 IL-6对chemerin mRNA表达的负调控可能是其引起胰岛素抵抗和糖脂代谢紊乱的机制之一,chemerin可能是噻唑烷二酮(TZD)类药物改善胰岛素敏感性的靶基因.
