摘要
[目的]建立离体跳动灌流心房模型,并探讨激活M2型毒蕈碱受体对离体跳动灌流心房钠尿肽(ANP)分泌的影响.[方法]利用急性剥离的家兔左心房和放射免疫分析方法观察M2型毒蕈碱受体对离体跳动灌流心房动力学及灌流液中ANP分泌的影响.[结果]记录到的离体跳动灌流心房模型心房内压及每搏搏出量与文献报道基本一致;卡巴胆碱明显抑制心房内压和搏出量,显著增加灌流心房ANP的分泌,此效应被M2型毒蕈碱受体抑制剂Methoctramine阻断.[结论]成功建立离体跳动灌流心房模型;M2型毒蕈碱受体的激活增加心房ANP的分泌.
OBJECTIVE To establish the perfused beating rabbit atria in vitro and to study the roles of activation of M2 muscarinic receptor for the atrial natriuretic peptide(ANP) secretion.METHODS The effects of M2 muscarinic receptor for perfused rabbit atria dynamics and ANP secretion in perfusate were observed by using the acute isolated left atrium of rabbits and radioimmunoassay.RESULTS The experimental recored intra-auricular pressure and stroke volume in the perfused beating rabbit atria in vitro were basically coincidence with the reported literatures.The carbachol inhibited obviously the intra-auricular pressure and stroke volume,and increased ANP secretion which blocked by Methoctramine,an inhibitor of muscarinic acetylcholine receptor.CONCLUSION The perfused beating rabbit atria has successfully established,and activation of M2 muscarinic receptor increases ANP secretion in perfused beating rabbit atria.
