环磷酰胺、秋水仙碱对急性脑梗死大鼠感觉运动功能的影响

摘要 目的探讨环磷酰胺、秋水仙碱对急性脑梗死大鼠感觉运动功能的影响。方法采用改良的Zea-Longa法建立大鼠MCAOI/R模型,实验组大鼠(n=10)给予环磷酰胺、秋水仙碱;对照组大鼠(n=10)给予生理盐水。在大鼠MCAOI/R术后24h、第1周、第2周、第3周、第4周进行神经功能评分。4周后取脑组织行组织化学染色。结果 2组大鼠Bederson神经功能评分第1~4周差异有统计学意义(P<0.05);平衡木行走试验评分第1周差异无统计学意义(P>0.05),第2~4周差异有统计学意义(P<0.05);倾斜板试验评分第3~4周差异有统计学意义(P<0.05);肢体放置试验评分第1~4周差异均有统计学意义(P<0.05)。2组大鼠的MAP-2阳性表达差异有统计学意义(P<0.05)。GFAP的阳性表达2组间差异无统计学意义(P>0.05)。结论环磷酰胺、秋水仙碱能够提高急性脑梗死大鼠的运动感觉功能,为临床抗炎细胞治疗急性脑梗死提供实验依据。 Objective To investigate the influences of cyclophosphamide and colchicine on the sensorimotor function of acute cerebral infarction rats.Methods The models of middle cerebral artery occlusion ischemic/reperfusion(MCAO I/R) were established with the Zea-Longa suture method.Twenty rats were divided into 2 groups randomly: treatment group(n=10),the rats were given cyclophosphamide and colchicine;control group(n=10),the rats were given sodium chloride.The functional outcome was investigated in 24 hours,1 week,2 weeks,3 weeks,4 weeks after the operation.The positive expressions of MAP-2 and GFAP were investigated 4 weeks later after the MCAO I/R.Results Rats given cyclophosphamide and colchicine performed significantly better than control group in scores of Bederson neural function,beam walking test,inclined plane test and limb placement test.The positive expressions of MAP-2 in theat ment group were significantly better than those of control group.The positive expressions of GFAP were mild in the two groups.Conclusion Cyclophosphamide and colchicine can improve the sensorimotor function of MACO I/R rats.
