4种甘草黄酮类化合物抗氧化构效关系研究 被引量:18

摘要 目的通过测定甘草苷、甘草素、异甘草苷和异甘草素对体外6种氧自由基的清除能力探讨4种化合物抗氧化作用的构效关系。方法采用化学方法测定4种甘草黄酮类化合物对羟自由基,金属离子螯合能力,DPPH,超氧阴离子清除率,亚油酸过氧化抑制率和抑制脂质过氧化的抑制率,考察其抗氧化效果。结果 4种化合物对DPPH、金属离子螯合能力和超氧阴离子的清除能力从大到小依次为异甘草苷,异甘草素,甘草素,甘草苷;对羟自由基和卵黄脂质过氧化的抑制能力为异甘草素,异甘草苷,甘草素,甘草苷;对亚油酸过氧化的抑制能力为异甘草苷,异甘草素,甘草苷,甘草素。结论酚羟基是起抗氧化作用的必要基团,C环开环后形成的羟基和双键是其抗氧化活性显著增强的重要原因。 Objective To study the structure-activity relationship of liquiritin,liquiritigenin,isoliquiritin and isoliquiritigenin by determining their antioxidant activities.Methods The chemical methods were used to determine hydroxyl radical scavenging ability,DPPH scavenging ability,metal ion chelating ability,superoxide anion scavenging ability,linoleic acid peroxidation inhibition ability and lipid peroxidation inhibition ability.Results DPPH scavenging ability,metal ion chelating ability,superoxide anion scavenging ability of the four compounds were arranged as isoliquiritin,isoliquiritigenin,liquiritigenin and liquiritin.Hydroxyl radical scavenging ability and lipid peroxidation inhibition ability were arranged as isoliquiritigenin,isoliquiritin,liquiritigenin and liquiritin.Linoleic acid peroxidation inhibition ability were arranged as isoliquiritin,isoliquiritigenin,liquiritin and liquiritigenin.Conclusion Phenolic hydroxyl group is necessary for Chalcone antioxidant;Hydroxyl and double bonds of the opened C ring can increase antioxidant activity.
