摘要
目的以东北红豆杉侧枝及叶为原料,采用细胞破碎与回流提取相结合的方法提取紫杉醇并测定其含量。方法通过对细胞破碎-回流提取法的破碎时间、破碎功率、乙醇浓度等单因素实验确定最佳单因素水平并通过正交实验,确定了以乙醇为提取剂提取红豆杉中紫杉醇类化合物的最佳工艺条件。然后采用高效液相色谱法测定紫杉醇含量。结果最佳提取条件:细胞破碎-回流提取破碎功率为70 W,乙醇浓度为95%,细胞破碎时间为40 m in。高效液相色谱法测定紫杉醇回收率为86.22%~99.07%,精密度RSD为3.45%,重现性RSD为3.41%,红豆杉叶中紫杉醇含量为0.173 3 mg/g,红豆杉侧枝中紫杉醇含量为0.171 8 mg/g。结论采用细胞破碎-回流提取方法提取红豆杉中紫杉醇可较大提高提取效率;用高效液相色谱法测定紫杉醇含量方法准确、可行。
Objective To extract paclitaxel from northeast China's taxus with cell disrupture-backflow technology and to measure its content.Methods The optimal condition to extract paclitaxel from taxus was determined by orthogonal test method and the time,temperature and ethanol concentration for cell disrupture-backflow were investigated,respectively.The liquid chromatogram was used to test the content of paclitaxel.Results The optimal extraction conditions were 5h,95% ethanol concentration and 100℃ for cells disruprure-backflow extraction,respectively.The results of liquid chromatogram were as follows: 92.28%(return rate of paclitaxel),3.45%(precision),3.41%(repeatability),1.733mg/g(the content of paclitaxel in taxus).1.718 mg/g(the content of branch in taxus).Conclusion Cells disrupture-backflow technology is able to improve the paclitaxel extraction efficiency from the taxus.Moreover,the content of paclitaxel in taxus can be mensured by liquid chromatogram.
