湖州雪藕的茎尖离体培养技术研究

Study on the Isolated Culture Technique of Stem Apex of Huzhou Xueou

[目的]为莲藕种藕的快速繁殖提供参考。[方法]以湖州主栽莲藕品种"迟来早"的地下茎尖为外植体,考察不同消毒时间和消毒方式(0.1%HgCl2、1.0%NaClO+0.1%HgCl2)对外植体的消毒效果,并研究取材时间和培养基中6-BA、NAA浓度对外植体分化的影响。[结果]HgCl2和NaClO联合对外植体消毒效果最好;最佳诱芽培养基为MS+1.0 mg/L6-BA+0.2 mg/L NAA;莲藕地下茎处于冬季休眠期时取材最佳,外植体分化率最高;将再生芽接种到培养基MS+1.0 mg/L6-BA上进行增殖,然后挑选生长良好的芽苗接种到生根培养基MS+0.1 mg/L6-BA+1.0 mg/LIBA+0.2%AC上可正常生根。[结论]该研究确定了莲藕品种"迟来早"地下茎尖的最佳离体培养条件。
