丙戊酸钠抑制胃癌细胞系-BGC823增殖的实验研究

摘要 目的探讨丙戊酸钠抑制胃癌细胞系BGC-823增殖作用。方法将0.75mmol/L^4.0mmol/L丙戊酸钠作用于BGC-823细胞48小时,观察细胞数量、形态的变化并用MTT法分析细胞生长抑制、AnnexinⅤ/PI双染法检测细胞凋亡率的改变和PI法观察细胞周期的变化。结果1.丙戊酸钠干预后细胞数量明显减少、形态不规则、细胞核固缩、胞浆减少;2.与对照组比较,丙戊酸钠可造成BGC-823细胞生长抑制率(F=417.11,p=0.000)、细胞凋亡率(F=385.24,p=0.000);以及增殖周期G1期比例(F=385,p=0.000)明显升高。结论丙戊酸钠可明显抑制HepG2的生长,诱导凋亡和细胞周期G1期阻滞作用,在胃癌治疗方面具有重要理论价值和实践意义。 Objective To study the depressant effect of Valproate acid sodium(VPA) on the proliferation of human gastric cancer cell line,BGC-823.Methods BGC-823 cells were treated with 0.75~4.0mmol/L VPA for 48 hours respectively;The cell growth condition was observed by invert microscope and inhabitation rate was studied by MTT assay;The apoptosis and cell cycle were analyzed by flow cytometry with AnnexinⅤ/PI and PI assay.Results The population decreased and shape,size changed after VPA treating;The inhabitation rate,apoptosis rate and the percentage of G1 phase in cell cycle of BGC-823 which treated with 0.75mmol/L~4.0mmol/L VPA increased significantly after 48 hours.Contrast to control group,The inhabitation rate(F=417.11,p=0.000),apoptosis rate(F=385.24,p=0.000) and percentage of G1 phase(F=385,p=0.000) changes are significant.Conclusion VPA can significantly suppress the growth of BGC-823 and induce apoptosis,cell cycle arrest in G1 phase effectively.
