摘要
目的观察GnRHα长周期辅助超排卵对小鼠子宫内膜组织αv、β1表达的影响,探讨提高IVF-ET妊娠率的方法。方法将36只小鼠随机分成三组,第一组生理盐水(NS)组;第二组HMG+HCG;第三组GnRHα+HMG+HCG。第二、第三组注射HCG24、48h和第一组观察排卵后24h、48h,采用免疫组织化学技术检测子宫内膜整合素av、β1的表达。结果整合素αv、β1在第一组呈现强阳性表达,第三组表达稍弱了第一组,第三组弱于前两组,第一、第二组两者比较无显著差异(P>0.05);第三组与前两组比较有显著差异(P<0.05)。结论控制性超排卵(COH)药物GnRHα可以影响小鼠分泌期子宫内膜αv、1β的表达,从而影响子宫内膜容受性,导致临床妊娠率偏低。
Objective To study tile effect of GnRHα assisted controlled ovarian hyperstimulation on the expression of integrins αv and β_1 in Mouse and the strategies to increase the rate of pregnancy in IVF-ET.Methods Thirty-six mice were divided into three groups randomly.Group one(NO.1) were perform abdomen injection with 0.9% salt solution(group NS),Group two with(NO.2) HMG+HCG and group three(NO.3) with GnRHα +HMG+HCG The thickness of endometrium were determined by computerized multi-thnctional imageandyzer NO.1 were detected 12h,48h after ovulation,while NO.2 and NO.3 24 and 48 hours after injection of HCG The expression of integrins αv and B 3in secretion endometrium of mouse,was examined by immunohistochemistry techniques.Results more stronger expressions of Integrins αv and β_1 were found in Group One and Goup Two than in Group three.However,in Group Three they were expressed obviously lower than the former two groups,the differences among them were significant(P<0.05).Conclusion The treatment of Controlled Ovarian Hypemtimulation can inhibit the expressions of Integrins αv and β_1 in Secretion Endometrium of mouse.Using GnRHα in the period of controlled ovarian hyperstimulation may reduced the endometrial receptivity and further the rate of pregnancy.However.it has no effectiveness on the thickness of endometrium.
