用组织芯片技术检测Sema4C基因在小鼠不同组织的表达水平

摘要 目的:观察在小鼠不同组织中的表达水平。方法:用地高辛进行标记并制备Sema4C特异性RNA探针对小鼠组织芯片进行原位杂交。结果:Sema4C在成年小鼠的大脑皮层,小脑蒲肯野纤维层,视网膜视神经节细胞层和肾小管上皮细胞层中均有较高表达.而在肌肉,心脏.肺,脾脏.肝脏等组织中表达量比较低或未检测到表达。结论:Sema4C基因主要表达在神经系统和肾脏,这将有助于进一步认识Sema4C功能。 Objective:To determine tbe extent of Sema4C expression in different tissues of mice.Methods:The specific Sema4C RNA probe was labelled by DIG,which was then hybridized with different tissues of mice ar- ranged in tissue microarray.Results:Sema4C was highly expressed in different tissues,including cortex of cere- brum,Purkinje's cells of cerebellum,optic nerve cells and the epithelium of renal fubules nephridium,but relative- ly low or little expressed in muscle,heart,lung,spleen and liver.Conclusion:Sema4C gene was mainly expressed in nervous system and kidney,the finding might be helpful in further exploring the potential function of Sema4C.
