骨髓间充质干细胞移植对急性肺损伤大鼠血浆TNF-α、IL-6水平的影响

Effects of bone marrow mesenchymal stem cell transplantation on plasma TNF-α/IL-6 level in acute lung injury in rat

目的：探讨骨髓间充质干细胞(MSCs)移植对急性肺损伤大鼠血浆TNF-α及IL-6水平的影响。方法：采用内毒素攻击法建立大鼠急性肺损伤模型,经静脉移植体外扩增培养的异体成年大鼠骨髓MSCs,检测伤后2h、4h、6h大鼠血浆TNF-α及IL-6水平的变化及对肺组织病理改变。结果：与单纯损伤组相比,MSCs移植能降低内毒素肺损伤大鼠血浆TNF-α及IL-6水平,降低肺湿/干重比值、髓过氧化酶(MPO)活性,减轻肺泡炎症损伤。结论：骨髓MSCs移植对急性肺损伤大鼠具有保护作用。

Objective:To evaluate the effect of mesenchymal stem cell transplantation on plasma TNF-α/IL-6 ex- pression in acute lung injury in rat.Methods:Acute lung injury model was rebroduced by intravenous in iection in travenous injured of LPS.Rat MSCs were isolated and cultured in vitro and transplanted into rats.The levels of plasma TNF-α/IL-6 were determined at 2h,4h and 6h after LPS administration.Results:In comparison with placebo group,plasma cytokine TNF-α/IL-6 expression was suppressed and lung tissue wet/dry ratio and MPO activity lowened at different time points after lung injury.Conclusion:MSCs therapy shows protective effects in LPS against lung injury.
