机械牵张应力对大鼠肺泡巨噬细胞CD_(14)表达的影响

摘要 目的观察机械牵张应力对大鼠肺泡巨噬细胞(PAM)CD_(14)表达的影响,并探讨其意义。方法将10~12周雄性SD大鼠放血处死,收集支气管肺泡灌洗液,分离、培养PAM,传至2代后接种于6孔培养板中培养48 h。将PAM随机分为5组,S1~S4组采用Flexercell 4000^(TM)应力加载系统分别施加5%、10%、15%和20%的机械牵张应力,频率30次/min、时间为24 h。对照组不处理。分别采用免疫组化和RT-PCR法测定各组大鼠肺泡PAM中的CD_(14)蛋白及mRNA。结果 S2~S4组CD_(14)蛋白及mRNA表达均显著高于对照组及S1组,尤以S3组、S4组为著(P均<0.05)。结论机械牵张应力可上调PAM表面CD_(14)表达。 Objective To investigate the effects of different mechanical stretch on on CD_(14) expression in alveolar macrophage (AMs) in rat.Methods 6 male SD rats were anesthetized and sacrificed by bloodletting.The lungs were lavaged with macrophage isolation buffer.Lavage samples were centrifuged and the AMs were resuspended in RPMI1640.After being cultured for 48 h,AMs were randomly divided into 5 groups(n = 6 each) and subjected to 0(group C),5%(group S1), 10%(group S2),15%(group S3) and 20%(group S4) elongation ...
