雌性大鼠去势后下丘脑视上核一氧化氮合酶阳性神经元的表达 被引量:2

摘要 目的:观察雌性大鼠行卵巢切除术后下丘脑视上区反映动物功能状态的一氧化氮合酶阳性神经元形态和数量的变化。方法:实验于2003-07/2004-01在山西医科大学神经解剖学科研室进行。将20只Wistar雌性大鼠随机分为两组:对照组和卵巢切除术后组,每组10只。将大鼠麻醉后取完整脑组织,制备冰冻切片,在光学显微镜下对两组大鼠下丘脑视上核的一氧化氮合酶阳性神经元进行形态观察和细胞计数,采用图像分析系统测定一氧化氮合酶阳性神经元免疫反应产物的平均灰度值。结果:20只大鼠均进入结果分析。①大鼠下丘脑视上区的一氧化氮合酶阳性神经元分布和形态观察:对照组分布广泛,以视上核的一氧化氮合酶阳性神经元表达较强,大量胞体呈椭圆形,密集成簇。卵巢切除术后组视上核一氧化氮合酶阳性神经元数目显著增多,胞体的截面积显著增大,轮廓很清晰;胞体上发出的突起粗且长,有个别纤维可见到明显的髓鞘。②下丘脑视上核一氧化氮合酶阳性神经元数目:卵巢切除术后组明显多于对照组犤(18.00±4.09)个/视野,(9.00±3.15)个/视野,t=5.514,P<0.0001犦。③下丘脑视上核一氧化氮合酶阳性产物的平均灰度值:卵巢切除术后组与对照组基本接近犤82.00±10.48,90.00±3.15,(t=0.985,P>0.5)犦。结论:雌性大鼠卵巢切除术后下丘脑视上核一氧化氮合酶阳性神经元数目反应性增多。 AIM: To observe the changes in the form and number of nitricoxide synthase (NOS) neurons, which reflect the animal functional status,in the supraoptic region of hypothalamus of female rats after ovariectomy. METHODS: The experiment was conducted in the Neuroanatomy Laboratory of Shanxi Medical University from July 2003 to January 2004. Twenty Wistar female rats were divided into 2 groups at random: One was the control group and the other was the post-ovariectomy group with 10 rats in each group. Intact brain tissues of the rats were gained to make frozen section after anesthesia. The formation observation and cell number of NOS positive neuron were detected in supraoptic nucleus of hypothalamus of rats in the two groups under light microscope. The average gray value of immunologic reaction product of NOS positive neuron was detected with image analysis system. RESULTS: Totally 20 rats were involved in the result analysis. ① The distribution and formation observation of NOS positive neurons in supraoptic nucleus of hypothalamus of rats: It was distributed widely in control group. The NOS neurons in supraoptic nucleus expressed strongly and a large number of cell body showed ellipse with intensive cluster. The number of NOS neurons in supraoptic nucleus significantly increased in post-ovariectomy group, and the sectional area of cell body became significantly bigger with clear outline; The apophysis on cell body was wide and long, and myelin sheath of some fiber could be seen. ② The number of NOS positive neurons in supraoptic nucleus of hypothalamus: It was significantly more in post-ovariectomy group than that in control group [(18.00±4.09),(9.00±3.15) per sight, (t=5.514, P 〈 0.000 1 )]. ③ The average gray value of NOS positive neurons in supraoptic nucleus of hypothalamus: It was similar in post-ovariectomy group and control group (82.00±10.48,90.00±3.15,t=0.985,P 〉 0.5). CONCLUSION: The reactivity of NOS positive neurons in supraoptic nucleus of hypothalamus in creases in female rats after ovariectomy.
