摘要
采取CaCl2-HOAc提取法提取样品中淀粉,探讨了光度法测定其含量的最优条件.试验结果表明,在pH 3.0~3.2介质中,标准可溶性淀粉溶液浓度Co与标准样品淀粉溶液浓度Cs的关系为:Cs=1.514 5 Co+0.008.并对不同食物样品进行淀粉含量测定,回收率为95.0%~99.0%之间.
Starch was extracted from foodstuffs with CaCl2-HOAc and absorbance determined in form of iodine-starch at wavelength of 570 nm. Working conditions were optimized. Experimental results showed that in a medium of pH 3.0--3.2, a correction relationship exists between soluble starch and the standard Chinese sorghum,as in the equation Cs = 1. 514 5 C0 +0. 008, Cs was the concentration (g · L^-1) of Chinese sorghum and C0 the concentration (g·L^-1) of soluble starch solution. The method has been applied to the determination of starch in various foodstuffs with recoveries of 95.0%--99.0%.
