反义bcl-2寡核苷酸对半乳糖诱导的晶状体上皮细胞中Bcl-2蛋白及细胞周期的影响

摘要 目的:探讨反义bcl-2寡核苷酸(bcl-2 ASODN)对半乳糖诱导的兔晶状体中Bcl-2蛋白表达及细胞周期的影响.方法:人工合成与bcl-2基因翻译起始区序列互补的寡核苷酸并磷酸化修饰,用半乳糖和bcl-2 ASODN处理体外培养的兔晶状体,流式细胞术检测bcl-2 ASODN对半乳糖诱导的LEC中Bcl-2蛋白表达水平及细胞周期的影响.结果:5.0~15.0μmol/L反义bcl-2寡核苷酸能下调半乳糖诱导的晶状体上皮细胞中Bcl-2蛋白的表达,荧光强度从158.70±2.14降至113.94±2.88,同时可降低S期细胞数,从20.60%降至0.67%.结论:bcl-2 ASODN能抑制半乳糖诱导的晶状体中Bcl-2蛋白的表达水平,阻止细胞进入S期,抑制细胞增殖. AIM: To investigate the effect of antisense oligodeoxynucleotide bcl-2 (bcl-2 ASODN) on the expression of Bcl-2 protein and the cell cycle of rabbit lens epithelial cells (LECs) induced by galactose, METHODS: From translation initiation region of bcl-2 gene, oligonucleotides were synthesized and modified with phosphorothioate. Rabbit lens were treated with galactose and bcl-2 ASODN. The influence of bcl-2 ASODN on the level of Bcl-2 protein expression and the cell cycle of LECs induced by galactose was detected by flow cytometry. RESULTS: 5. 0 - 15.0 μmol/L of bcl-2 ASODN reduced the level of Bcl-2 protein expression on LECs induced by galactose, with the fluorescence intensity decreasing from 158.70±2. 14 to 113.94 ± 2.88, and decreased the number of S phase cells of Lens from 20.60% to. 0. 67%. CONCLUSION: bcl-2 ASODN can inhibit the level of Bcl-2 protein expression on LECs induced by galactose, interfere the entry of LECs into the S phase and inhibit the proliferation of LECs.
