摘要
采用共价-偶联法,在较温和的条件下进行漆酶在Fe3O4/SiO2载体上的固定化研究,利用TEM、FTIR、UV-vis和VSM等手段对固定化漆酶进行表征并考察其酶学性质.结果表明,漆酶在Fe3O4/SiO2载体上固载之后,其最适pH和温度变化不大,但操作稳定性和分离性能都有明显改善,以ABTS为底物,固定化漆酶在重复使用10次后,酶活仍能保持在初始酶活的85%以上,反应之后的漆酶可以通过外加磁场快速分离.
The immobilization of laccase from Trametes Versicolor on magnetic Fe3O4/SiO2 support by cross-linking method under mild conditions is investigated.TEM,FTIR,UV-vis and VSM measurements are used to characterize the immobilized laccase.The results show that the immobilization of laccase has little effect on optimum pH and temperture,while thermal stability,operation stability and separation are improved greatly.Using ABTS as substrate,the immobilized laccase is tested and separated under an external magnetic field,after 10 cycles of repeated use,the activities of enzyme still maintaine above 85% of the intial activity.
