早产、足月胎兔动脉导管平滑肌细胞中胱硫醚-γ-裂解酶mRNA表达及意义 被引量:1

Expression and Significance of Cystathionine-γ-lyase mRNA in Dctus Arteriosus Smooth Muscle Cells of Fetal Rabbits

摘要 目的:研究早产和足月胎兔动脉导管平滑肌细胞胱硫醚-γ-裂解酶(cystathionine-γ-lyase,CSE)mRNA的表达情况。方法:选取日本大耳白胎兔早产组(孕25 d)、足月组(孕30 d)各10只,分离动脉导管平滑肌组织,采用组织块改良贴壁法进行胎兔动脉导管平滑肌细胞的体外培养,半定量逆转录集合酶链(RT-PCR)检测胱硫醚-γ-裂解酶mRNA的表达。结果:经10~15 d培养后,倒置显微镜下观察,动脉导管平滑肌细胞呈梭形或放射状生长,高低起伏形成典型的"峰-谷"样,早产组和足月组CSE mRNA的表达值分别为(29.67±6.7)和(10.35±4.5),两组比较差异有统计学意义(P<0.05)。结论:早产及足月胎兔动脉导管平滑肌细胞均表达CSE mRNA,表达量可能与胎龄有关,提示动脉导管组织可能内源性生成H2S气体,且在宫内低氧环境中H2S可能是维持动脉导管开放状态的一个重要因素。 Objective To investigate the expression of cystathionine-γ-lyase(CSE) mRNA in ductus arteriosus smooth muscle cells of premature and full-term fetal rabbits.Methods The ductus arteriosus were obtained from premature fetal rabbit group with 25 days gestation and full-term fetal rabbit group with 30 days gestation(n=10),respectively.The smooth muscle cells were primary cultured with modified attached method in vitro,the expression of cystathionine-γ-lyase mRNA were determined with RT-PCR.Results After cultured for 10 to 15 days,the cells were grown as fusiform and radiate form under microscope.The expression of cystathionine-γ-lyase mRNA in premature group(29.67±6.7) was significantly higher than that of full-term group(10.35±4.5),(P<0.05).Conclusion The expression of CSE mRNA were observed with a gestational age dependent manner in both groups,which demonstrate that H2S could endogenous create in ductus arteriosus smooth muscle cells and H2S may be an important factor in maintenance opening of ductus arteriosus.
