赖氨酰氧化酶对荷胃癌裸鼠肿瘤形成和转移的影响

Effect of Lysyl Oxidase on Tumorigenesis and Tumor Metastasis in Gastric Tumor-bearing Nude Mice

摘要 目的观察用β-氨基丙腈(β-aminopropionitrile,BAPN)抑制赖氨酰氧化酶(lysyl oxidase,LOX)对荷胃癌裸鼠肿瘤形成和转移的影响。方法 6周龄的裸鼠,随机分为BAPN预处理组和对照组,预处理组10只,对照组15只。BAPN预处理组每天用BAPN 100mg.kg-1腹腔注射,对照组每天用相同体积的无菌PBS腹腔注射。2周后,以人胃癌细胞SGC-7901 1×107.mL-1建立荷胃癌裸鼠模型。肿瘤细胞接种4周后,比较成瘤率、瘤重和转移率。结果 BAPN预处理组平均瘤重高于对照组,P<0.05;BAPN预处理组的成瘤率和转移率与对照组的差异无统计学意义。结论用BAPN预先抑制LOX活性后,肿瘤的重量明显增加,但对原发肿瘤形成和转移未见明显作用。 Objective To observe effect of lysyl oxidase(LOX) inhibited by β-Aminopropionitrile(BAPN) on the tumorigenesis and tumor metastasis in gastric tumor-bearing nude mice.Methods 25 Nude mice were randomly divided into two groups,the group of pre-treat with BAPN(10 mice) and control group(15 mice).The mice in pre-treat group were peritoneal injection with 100 mg.kg-1 BAPN every day,and the mice in control group were treated with PBS in the same volume every day.Two weeks later,all mice were injected subcutaneously with gastric cancer cells to form models.Four weeks after the cancer cells were injected,all mice were sacrificed and the rate of tumorigenesis,the rate of tumor metastasis and the weight of tumor were compared between groups.Results The weight of tumor in pre-treat group was higher than that in control group,(P < 0.05).The rate of tumorigenesis and metastasis had no statistically significant difference between groups.Conclusion Inhibition of LOX by BAPN in advance increased the average weight of tumors.
