摘要
目的观察新风胶囊对佐剂性关节炎(adjuvantarthritis,AA)大鼠滑膜细胞凋亡的影响。方法以弗氏完全佐剂复制出大鼠AA模型,50只雄性Wistar大鼠随机分为正常对照组、模型对照组、甲氨喋呤(MTX)对照组、雷公藤多甙(TPT)对照组和新风胶囊(XFC)治疗组,各组10只。分别采用电子显微镜技术、免疫组化法及流式细胞术检测了各组大鼠滑膜细胞凋亡情况。结果电镜观察显示,新风胶囊组滑膜细胞核染色质呈凝块状,边聚呈月芽状,胞浆内及胞膜外可见凋亡小体;免疫组化法观察显示,与模型组比较,XFC组FasL的表达显著增加,bcl2表达显著减少(P<0.05);与MTX组、TPT组比较,XFC组上述指标变化差异均无显著意义(P>0.05)。流式细胞术观察表明,XFC组滑膜细胞凋亡率显著高于模型组、TPT及MTX组(P<0.05)。结论新风胶囊通过促进滑膜细胞凋亡,抑制滑膜增生,从而消除关节滑膜肿胀,降低关节炎指数。
Objective To observe the effect of Xinfeng capsule(XFC)on the apoptosis of the synoviocyte of adjuvant arthritis (AA) rat. Methods Model rats were established by the Freund's complete adjuvant. Fifty rats were randomly divided into 5 groups: normal control, model control, methotrexate (MTX) control, triperygium wilfordii polycoride tablet (TPT) control,xinfeng capsule treated (XFC). the apoptotic manifestation of the synoviocyte was observed by electron microscopy, immunohisto chemistry (IHC) and flow cytometry (FCM). Results Observed by TEM:The synoviocyte and thymus-lymphocyte in XFC group was wizened, their chromatin of the nuclear presented the changing of gelosis and shape of “crescent moon”. Results by IHC: Compared with the model group, FasL expression of synoviocyte was increased in the XFC group significantly (P〈 0.05 ), but bcl-2 expression decreased, Fas expression showed significant difference ( P 〉 0.05 ). The gene expressions between the three groups treated showed no significant difference ( P 〉 0.05). Results examined by FCM : the apoptotic percentage of synoviocyte in XFC group was higher than those of TPT and MTX group significantly ( P 〈 0.05). Conclusion XFC can eliminate the swelling of joint and reduce AI by promoting the apoptosis of sgnoviocye and inhibiting the hyperplasia of synoviocyte.
