摘要
以MS为基本培养基,通过调节6-芐氨基嘌呤(6-BA)、3-吲哚丁酸(IBA)、1-萘乙酸(NAA)、3-吲哚乙酸(IAA)等4种生长素的配比和浓度,成功地用组织培养的方法获得辽东楤木茎尖、茎等部位诱导产生的愈伤组织和再生植株,结果表明:辽东楤木离体培养的愈伤组织诱导、分化成苗和分化生根的最佳激素配比为NAA0.01mg·L-1+6-BA0.5mg·L-1、NAA0.02mg·L-1+6-BA0.5mg·L-1和NAA0.01mg·L-1+IBA1.5mg·L-1,应用该技术体系的愈伤组织诱导率、分化率和再生植株成活率分别为94%、22.5%、98%,该技术体系为国内外同类研究的首次报道。
Japanese Aralia tissue culture on MS medium. regulated with the combination and concentration of 6-BA,IBA,NAA and IAA, was studied. The regenerating plantlets were successfully obtained. The plant organ of shoot tip, stem could be induced to produce callus and regenerate seedlings in vitro culture system, The best combinations of concentrations with hormone for callus inducing, regenerated bud and root inducing are NAA0.01mg·L^-1+6-BA0.5mg·L^-1, NAA0.02mg·L^-1+6-BA0.5mg·L^-1and NAA0.01mg·L^-1+IBA1.5mg·L^-1. The percentage of the callus inducement,seedling differentiation and plantlet survival after seedling cultivating were 94%,22.5% and 98% respectively.This in vitro culture system of Japanese Aralia was the first report so far.
